چکیده

Two human erythroleukemia cell lines, HEL and KS62, express transglutaminase activity. The enzyme was identified as a tissue transglutaminase following Chromatographie purification. All-fra/w-retinoic acid (10 MM)stimulated differentiation in HEL cells as judged by a 4-fold increase in hemoglobin content and a reduction in cell proliferation. The transglutaminase activity increased 9-fold. This increase in transglutaminase was the result of a pretranslational regulation of the gene as revealed by Northern blot analysis of inKNV. These changes were not a result of cell apoptosis, since parallel DNA degradation catalyzed by a ( V ' -dependent endonuclease could not be demonstrated. The K562 cells, in contrast, showed no transglutaminase induction following exposure to retinoic acid and displayed no changes in maturation markers or cell growth.

Analysis of Promyelocytic Leukemia in Human Embryonic Carcinoma Stem Cells During Retinoic Acid-Induced Neural Differentiation

Background: Promyelocytic leukemia protein (PML) is a tumor suppressor protein that is involved in myeloid cell differentiation in response to retinoic acid (RA). In addition, RA acts as a natural morphogen in neural development. Objectives: This study aimed to examine PML gene expression in different stages of in vitro neural differentiation of NT2 cells, and to investigate the possible role o...

Evaluation of Neurogenic Potential of Human Umbilical Cord Mesenchymal Cells a Time- and Concentration- Dependent Manner

Background: Retinoic acid as one of the most important regulators for cell differentiation was examined in this study for differentiation of human umbilical mesenchymal cells (hUCM). Methods: After isolation, hUCM were evaluated for mesenchymal stem cell properties by flow cytometry and alkaline phosphatase assay. Also, doubling time of the cells and their differentiation potential into adipoge...

تاثیر رتینوئیک اسید در تمایز سلّول‎های بنیادی مزانشیمال بافت چربی به سلّول‎های زایا

Background: Recent publications regarding the differentiation of stem cells to germ cells have motivated researchers to make new approaches in infertility. In vitro production of germ cells improves the understanding of differentiation process of male and female germ cells. Since using embryonic stem cells for this purpose has been associated with tumorogenesis and ethical criticisms, the men...

A New Two Step Induction Protocol for Neural Differentiation of Human Umbilical Cord Blood-Derived Mesenchymal Stem Cells

Background: In this study, we examined a new two step induction protocol for improving the differentiation of human umbilical cord blood-derived mesenchymal stem cells into neural progenitor cells. Materials and Methods: Human umbilical cord blood-derived mesenchymal stem cells were first cultured in Dulbecco’s modified eagle medium supplemented with 10% fetal bovine serum in a humidified incu...
